Deschloroetizolam
Deschloroetizolam is a thienotriazolodiazepine and the dechlorinated analog of etizolam.1 It first appeared through grey market research chemical vendors in 2014 and is not prescribed for any medical use. It produces depressant, anxiolytic, sedative, muscle relaxant, and amnesic effects through modulation of GABA receptors. Compared to its parent compound etizolam, it is roughly half as potent but exhibits approximately twice the duration of action.

Paradoxical reactions to benzodiazepines such as increased seizures (in epileptics), aggression, increased anxiety, violent behavior, loss of impulse control, irritability and suicidal behavior sometimes occur (although they are rare in the general population, with an incidence rate below 1%). These paradoxical effects occur with greater frequency in recreational abusers, individuals with mental disorders, children, and patients on high-dosage regimes.

Deschloroetizolam acts as a positive allosteric modulator at GABA-A receptors via the benzodiazepine binding site.citation needed By enhancing the efficiency of the inhibitory neurotransmitter gamma-aminobutyric acid (GABA) at these receptors, it produces its characteristic depressant and anxiolytic effects. It is the dechlorinated analog of etizolam and is reported to be approximately half as potent as that parent compound.2

Limited pharmacokinetic data is available for deschloroetizolam.3 It is reported to have a bioavailability similar to that of its parent compound etizolam.
