4-MeO-PCP is an arylcyclohexylamine synthetic dissociative that can produce hallucinogenic and anesthetic effects. First synthesized in 1965 by Parke-Davis chemist Victor Maddox, it became the first arylcyclohexylamine research chemical sold online when it appeared in late 2008. It is reported to have reduced potency relative to PCP. Dosage information remains tentative, potentially due to variations in the potency of available supplies.

Pharmacodynamics

4-MeO-PCP acts primarily as an NMDA receptor antagonist.1 Its NMDA affinity is lower than that of PCP but higher than that of ketamine. The compound also binds with notable affinity to σ1 and σ2 receptors, and shows moderate affinity for the norepinephrine and serotonin transporters.1 It is considered a relatively selective NMDA receptor ligand, lacking appreciable affinity for the dopamine transporter.1

Pharmacokinetics

4-MeO-PCP is orally active, indicating sufficient oral bioavailability for pharmacological effects. There have been no studies on the pharmacokinetic profile of 4-MeO-PCP and the metabolism of 4-MeO-PCP is unknown.

Repeated excessive use over extended periods may cause bladder and urinary tract problems similar to those seen with ketamine, including urinary frequency and urgency, pelvic and bladder pain, hematuria, and incontinence; these effects can be avoided by limiting use to less than weekly.

Psychosis Risk

Psychotic episodes, disturbing hallucinations, delusions, anxiety, and paranoia are reported as potential negative effects. Risk of adverse psychological reactions increases with higher doses and redosing.

4-MeO-PCP was originally synthesized in 1965 by Victor Maddox, a medicinal chemist working at the pharmaceutical company Parke-Davis. The compound remained largely obscure for decades following its initial creation.
