1P-ETH-LAD is a semisynthetic psychedelic of the lysergamide class and a suspected prodrug of ETH-LADcitation needed, structurally related to both LSD and 1P-LSD. Developed as a designer drug, it is reported to be moderately to significantly more potent than LSD. Its effects are often described as more visual and synaesthetic, with a deeper, less emotionally-charged headspace. It remains largely unresearched and is classified as a research chemical.

Pharmacodynamics

Direct receptor pharmacology of intact 1P-ETH-LAD has not been measured; the located 1P-ETH-LAD study was an analytical and serum-incubation study rather than a receptor assay.citation needed Formation of ETH-LAD in human serum supports a possible prodrug mechanism, consistent with findings reported for other 1-acyl-substituted LSD derivatives, but it does not establish that ETH-LAD alone mediates the effects of 1P-ETH-LAD in humans.1

Pharmacokinetics

In an in-vitro experiment, 1P-ETH-LAD was incubated in human serum at 37 °C and sampled hourly from 0 to 6 hours, with an additional sample at 24 hours. ETH-LAD was detectable at the first one-hour sampling point, while parent 1P-ETH-LAD remained detectable at 24 hours.2 These findings support possible in-vivo prodrug conversion but do not establish human bioavailability, conversion fraction, clearance, plasma half-life, or clinical effect duration.2

1P-ETH-LAD can produce tolerance very quickly after administration. This rapid buildup is typical of serotonergic psychedelics that act at the 5-HT2A receptor.3
Baseline Reset
Physiological tolerance typically returns to baseline after approximately 7 days of abstinence. Some users suggest that full restoration of the psychological or experiential aspects of the substance may require two weeks or longer between uses.

Approximately 3 days after ingestion, physiological tolerance is estimated to decrease to roughly half of its peak level.
