Addiction & Dependence
Psychological
ModerateAnimal studies demonstrate addictive potential, with rats learning to lever-press for injections into the nucleus accumbens and frontal cortex. Dizocilpine produces conditioned place preference and enhances self-stimulation responding, indicating reinforcing properties independent of dopamine pathways.
Toxicity
A fatal polydrug case has involved dizocilpine together with benzodiazepines and alcohol. Its high potency makes accurate dosing difficult, increasing overdose risk.
Administration induces the formation of brain lesions known as Olney's lesions in the posterior cingulate and retrosplenial cortex; vacuoles begin forming within 30 minutes of a 1 mg/kg subcutaneous dose in rats, with subsequent neuronal necrosis and glial response.
Psychosis Risk
Frequently used in animal models to mimic psychosis due to its pronounced psychotomimetic effects. A single injection successfully models both positive and negative symptoms of schizophrenia, while repeated administration produces neurochemical alterations similar to those found in first-episode schizophrenic patients. Chronic administration in laboratory animals resulted in neuropathological changes resembling those seen in schizophrenia.
