Flmodafinil acts as a selective, atypical dopamine reuptake inhibitor. It binds to the dopamine transporter (DAT) with a Ki of 4,090 nM1 and has been shown to block DAT by 83%citation needed. Its affinity for the serotonin transporter is approximately 12-fold lower than for the DAT, and it has negligible affinity for the sigma-1 receptor. In animal models, flmodafinil increases dopamine levels in the nucleus accumbens by up to 150 to 200% of baseline at the highest assessed doses.
