Flmodafinil acts as a selective atypical dopamine reuptake inhibitor, binding the dopamine transporter (DAT) with a Ki of 4,090 nM and blocking it by roughly 83%, a greater degree of blockade than methylphenidate without accompanying adrenergic effects. Its affinity for the serotonin transporter is around 12-fold weaker (Ki 48,700 nM), and it has negligible affinity for the sigma σ1 receptor (Ki > 100,000 nM). Both enantiomers raise dopamine levels in the nucleus accumbens in animals, reaching up to 150 to 200% of baseline at the highest doses tested, increases far smaller than those produced by amphetamine or cocaine. In animals the drug promotes wakefulness, maintaining it over a significantly longer timeframe than modafinil while perturbing sleep architecture to a lesser degree, with attenuated EEG power density in slow frequencies below 4 Hz and no sleep rebound.
