Chemical Class

5-APDB is a benzofuran-class entactogen and amphetamine stimulant with structure and effects similar to MDMA. It is an analogue of MDA in which a heterocyclic oxygen is replaced by a methylene bridge1. A team led by David E. Nichols at Purdue University investigated entactogen structure-activity relationships to determine whether potentially useful psychopharmacological properties could be dissociated from serotonergic neurotoxic effects1, and it was described in a 1993 Journal of Medicinal Chemistry paper1.

Pharmacology

Pharmacodynamics

5-APDB acts primarily as a highly selective serotonin releasing agent. It also weakly inhibits serotonin reuptake and inhibits dopamine and norepinephrine reuptake, with additional activity at serotonin receptors.

Psychological

Low

Classified as habit-forming. As an entactogenic stimulant with serotonin-releasing properties similar to MDMA, some potential for psychological habituation exists.

Psychosis Risk

Visual and auditory hallucinations have been reported as negative effects.

History & Culture

5-APDB was developed by a team led by David E. Nichols at Purdue University as part of research into non-neurotoxic MDMA analogues and was first described in 1993. Nichols and colleagues described 5-APDB and 6-APDB as MDMA analogues in that year.…
