HighChlordiazepoxide has significant abuse potential and is classified as habit-forming. Its presence is often found in urine testing from drug users without a prescription, suggesting misuse liability. The FDA requires boxed warnings for all benzodiazepines describing risks of abuse, misuse, and addiction.
Physical
HighChlordiazepoxide can produce physical dependence and benzodiazepine withdrawal syndrome. Its withdrawal profile resembles withdrawal from alcohol or barbiturates. Risk of unpleasant withdrawal increases with higher doses and longer use, but symptoms can still occur at standard doses and after short-term treatment.
Toxicity
Liver problems including yellow eyes and skin (jaundice) are listed among side effects; individuals with severe liver deficiencies such as hepatitis or cirrhosis show decreased elimination by a factor of two.
Laboratory tests found chlordiazepoxide produced toxicity in mouse spermatozoa, including abnormalities in both shape and size of sperm heads.
Chronic use leads to development of tolerance with a decrease in benzodiazepine binding sites in the forebrain; studies in mice showed impairment of latent learning, and chlordiazepoxide caused the most profound reduction in serotonin turnover among benzodiazepines tested.
Possible carcinogenesis and mutagenesis have been listed among adverse effects from benzodiazepine use during pregnancy.
